Sample Homogenization with the TissueLyser II
Before using the TissueLyser II for the first time, consult CCG staff for proper training. Additionally, review the manufacturer’s instructions.
Homogenization settings will vary considerably depending on sample type, preservation method, and the type of homogenization beads used. Some experimentation will be required to find the best method for your samples.
Quick start guide

Load the adapters with tubes or plates as required. Ensure samples are distributed evenly between shaker arms, and across each plate/tube holder.

Lift up the plastic hood covering the wiggle mechanism.

Ensure the point of the shaker clamp locking pins are lifted out of their grooves. Loosen the handwheels until the clamps are sufficiently loose to allow for insertion of the adapter assemblies. Note that when a locking pin is disengaged, its handwheel should loosen easily and will not click when tightened.
If a previously tightened handwheel will not loosen and its locking pin is disengaged, it may be necessary to tighten it slightly to allow its internal ratchet to disengage before loosening.

Insert the adapter sets, ensuring that the round “bumps” engage with the slots on the shaker arms.

Turn the locking pins so that their points fall into their grooves. Tighten the handwheels until the clamps hold the adapter assemblies securely. Note that when a locking pin is engaged, its handwheel will not loosen and will click when tightened.
Do not over-tighten the clamps; this can crush your tubes!

Close the plastic hood.

Power on the machine using the rocker switch on the rear of the machine by the power cord.

Press the PROG button to cycle through the machine’s presets. If a suitable preset exists, press START to run that preset. If no suitable preset exists, use the controls to adjust frequency and time as required.

When lysing is complete, remove the adapter assemblies (removal is the opposite of installation). Leave the machine powered down with clamps empty and hood closed.
